PsA is a highly inheritable condition. Approximately 40% of patients who develop PsA have a family member with either psoriasis or PsA. The genetic biomarker HLA-B27 is associated with joint involvement, particularly axial spondylarthritis. It is also associated with younger age of disease onset.

HLA-Cw6 has a strong association with cutaneous psoriasis, particularly early-onset psoriasis. There is minimal, if any, influence in the development of joint disease.

HLA-B22 and HLA-DR7 are protective markers.
